Job Overview We are seeking a Research Assistant with strong expertise in oligonucleotide synthesis, purification and quality control to support the development of the RNA Therapeutics Platform within the Sidney Altman Hub at the Montreal Clinical Research Institute (IRCM). The successful candidate will play a key role in establishing and operating oligonucleotide bioproduction and analytical capabilities for ASOs, siRNAs and sgRNAs. They will also help develop the platform’s therapeutic RNA and lipid nanoparticle activities in a highly collaborative environment, working closely with researchers, users and other team members. Responsibilities * Contribute to establishing and operating the RNA Therapeutics Platform in collaboration with the platform lead. * Perform chemical synthesis of therapeutic and research oligonucleotides, including ASOs, siRNAs, sgRNAs and other formats, using automated synthesis equipment, including solid phase synthesizers. * Perform oligonucleotide purification, desalting, lyophilization, quantification and characterization. * Conduct oligonucleotide quality control using analytical approaches such as HPLC, LC-MS, UV spectrophotometry and other relevant methods. * Assist with the maintenance, optimization and troubleshooting of oligonucleotide synthesis and analytical equipment. * Contribute to the development, optimization and standardization of synthesis, purification and quality-control protocols. * Maintain rigorous documentation of activities in accordance with applicable quality procedures. * Contribute to in vitro RNA synthesis, purification and quality-control activities. * Participate in lipid nanoparticle (LNP) formulation and characterization according to platform needs. * Support the implementation and maintenance of the platform’s quality-management systems. * Work with researchers and platform users to understand their needs, advise on experimental approaches and follow up on projects. * Contribute to the development of new services and the continuous improvement of platform processes and activities. * Participate actively in team meetings, knowledge sharing and continuous improvement of platform services and procedures. * Establish and maintain effective, collaborative working relationships with platform users and team members, and help deliver high-quality scientific services. What You Bring to the Table Essential Requirements * Bachelor (BAC) in chemistry, biochemistry, molecular biology, biotechnology or a related field. * Hands-on experience in chemical oligonucleotide synthesis. * Experience in oligonucleotide purification and quality control. * Experience with automated oligonucleotide synthesis equipment, such as Shasta, MerMade, Dr Oligo or equivalent. * Experience with analytical techniques such as HPLC, LC-MS, capillary electrophoresis or similar methods. * Knowledge of RNA synthesis and analytical methods is an asset. * Experience in lipid nanoparticle (LNP) formulation is an asset. * Knowledge of molecular biology and biochemistry, including PCR, cloning, electrophoresis, nucleic acid quantification and western blotting. Desired Skills * Excellent understanding of oligonucleotide chemistry and chemical modifications of nucleic acids. * Good knowledge of RNA biology and the mechanisms of action of therapeutic oligonucleotides. * Good understanding of quality-control principles applied to nucleic acids. * Ability to analyze and interpret complex analytical results. * Scientific rigour and excellent ability to document experimental activities. * Strong organizational skills and ability to manage multiple projects simultaneously. * Ability to work in a collaborative technology-platform environment. * Familiarity with laboratory information management systems (LIMS) and quality systems is an asset. * Knowledge of GMP/GLP is an asset. * Excellent teamwork skills in a multidisciplinary environment bringing together expertise in chemistry, biochemistry, molecular biology and RNA therapeutics. * Ability to communicate effectively with collaborators from varied backgrounds and contribute to a positive, engaging work environment.
